High throughput sequencing at ASGPB

Next Generation Sequencing (New)

ASGPB provides high throughput DNA sequencing services using the Roche Next Generation Sequencer GS FLX System (a.k.a. 454 Sequencer). The 454 GS FLX Titanium system uses bead based pyro-sequencing instead of Sanger dideoxy-termination sequencing and can obtain ~400 million nucleotides (bases) per run without any cloning biased.


Requirements

A minimum of 5 µg (for shotgun) or 15µg (for 8Kb Paired End) of high quality DNA (concentration and final volume depending on application) provided by the customers. ASGPB will prepare library, perform QC on library preparation and sequencing run.

Sanger Sequencing

The ASGPB genome infrastructure currently consists of several state-of-the-art capillary-based DNA sequencers and accessory equipment to deal with large sample volumes. The facility can process up to 9000 samples per day with a projected sequence output of 6.75 Million bases. Several prokaryotic and one eukaryotic genome project are currently in progress.

The facility offers DNA sequencing services for a wide range of templates including plasmid DNA, PCR fragments, cosmid DNA, and Bacterial Artificial Chromosomes (BACs). Sequencing reactions are performed using Applied Biosystems BigDye terminator chemistry and are run on ABI 3730XL capillary-based DNA sequencers.

Details

Turn-around time for sequencing is generally 1-2 days from receipt of the template and primers, depending on the workload. Additional services such as template preparation (plasmid preparation in 96-well blocks) will add to the turn-around time.

In addition, ASGPB offers the following services:

• Construction and validation of random-sheared DNA fragment libraries.
• High-throughput, automated template preparation using Eppendorff kits or the overnight high-throughputtemplate preparation via the Rolling Circle Amplification (RCA) method.
• Assembly and processing of DNA sequencing reactions in 96-well microtiter plate format.

ASGPB is committed to specifically support high-throughput projects such as genome-wide analyses and will assist with the preparation of relevant grant applications. In addition, ASGPB will provide assistance with sequence assembly and bioinformatic analysis of sequencing projects.
